Researchers uncovered that the tripartite motif-containing 63 (TRIM63) activates the AKT, p38, and ERK signaling pathways in facilitating the development of thyroid carcinomas and published their findings in Molecular and Cellular Endocrinology.
Thyroid carcinomas account for approximately 2.5% of all malignancies. The subtypes of thyroid carcinoma include papillary thyroid carcinoma (PTC), follicular thyroid carcinoma (FTC), and medullary thyroid carcinoma (MTC).
The tripartite motif family of proteins has recently been shown to play a role in regulating a number of cellular processes, such as proliferation, apoptosis, and autophagy. TRIM63, a member of the tripartite motif family, has been implicated in breast cancer, another endocrine malignancy. The authors of this study hypothesized that TRIM63 is likewise involved in thyroid carcinoma progression.

The research team first identified mRNA levels in selected thyroid carcinoma tissues and cell lines and studied the effects of modified TRIM63 on various cellular processes of thyroid carcinoma cells in vitro. They then established a xenograft model to investigate the functions of TRIM63 in vivo. Through this methodology, they were able to study the mechanisms of cancer progression aided by TRIM63.
The results demonstrated that by knocking down TRIM63, the protein expression of p-AKT, p-p38, and p-ERK in thyroid carcinoma cells was decreased. The opposite result could be observed when TRIM63 was overexpressed. Hence, the research team could be confident that TRIM63 plays a role in promoting the AKT, p38, and ERK signaling pathways.
They also discovered other ways in which TRIM63 supports the growth of thyroid carcinoma, but they chose to give weight to their findings on the AKT, p38, and ERK signaling pathways because they intend to investigate them further. The identification of the role that TRIM63 plays in driving the progression of thyroid carcinomas may open the door to the development of new forms of anticancer therapies.
